SuperFusion™ - Ultra Fidelity Taq DNA Polymerase
High-Fidelity DNA Polymerases are important for applications in which the DNA sequence needs to be correct after amplification. SuperFusion™ - Ultra Fidelity Taq DNA Polymerase possess high fidelity and faster processivity, therefore, the PCR process dramatically increases the yield of PCR products and the ability to amplify long templates in much shorter time. SuperFusion™ - Ultra Fidelity Taq DNA Polymerase possesses the 5'->3' DNA polymerase activity, 3'->5' exonuclease activities and temperature-depended strand-displacement activity and generates blunt ends in the amplification products.
Advantages
- Superior fidelity – about 50x improvement compared to Taq polymerase
- Excellent performance across a wide range of “difficult” templates Long range amplification of complex targets - > 12 kb from genomic DNA
- High speed - reduce reaction times Resistant to blood containing DNA samples (up to 30% of blood )
Applications
- High Fidelity (Hi-Fi) PCR
- “Hi–Fi” - LD PCR
- Direct PCR from Blood Specimens
Components
- SuperFusion™ Ultra Taq DNA polymerase
- 10x Buffer A: for fragments < 5kb
- 10x Buffer B: for fragments > 5kb

General Protocol
The optimal reaction conditions for SuperFusion™ - Ultra Fidelity Taq DNA Polymerase may differ from PCR protocols for standard (Taq-like) DNA polymerases. PCR conditions for SuperFusion™ - Ultra Fidelity Taq DNA Polymerase is very similar to PCR conditions like “Phusion-like” DNA polymerases, e.g. the enzyme works better at elevated denaturation and annealing temperatures. PCR reactions should be set up on ice. Prepare a master mix for the appropriate number of samples to be amplified.
Important Note
It is critical that the SuperFusion - Ultra Fidelity Taq DNA Polymerase is the last component to be added to the PCR master mix because the enzyme exhibits 3'- 5' exonuclease activity that can degrade primers in the absence of dNTPs.
